Sign Up Code - that's a great choice if you're giving the code out somewhere :)
...which is a likely scenario during testing.
However, for production, we work around this by modifying the registration form to read something like, "What color is the ocean?" And, of course, the signup code is "blue". Not wonderfully advanced, but it works.

I am using Anti-Splog. Seems to work very well with sign-ups on Buddypress and Multisite.

Also use the Signup Code plugin available here. It will add another layer of protection that you might want to continue using even after you go live.
