This is my first time to use WPMU. I've got a dedicated server and I THINK I would like to set up multiple directories such as /CMS1/, /CMS2/, etc. and upload a separate installation of WPMU to each directory for beta testing. In theory, I would then use a different theme in each version so I could just switch folders and compare how they look and function. Would this work?
Also, if it does work for beta testing and I find I like /CMS2/ and I want to make it live, can I do it with a simple redirect index.html on the main domain? If not, and I need to move it to the root, are the permalinks or anything else going to be messed up because of the move?

You can probably get away with it for beta testing but most of us usually recommend that you install mu within a separate hosting account or by itself in an active environment. I would stay away from any redirects as they usually cause additional strain on the server and folks have been booted for stuff like that. (Hi Andrea!)
I better idea for you may be to install a local server like XAMPP, set it up for the domain that you plan on using, set up mu locally, work on it and then upload it to your server when ready to roll it out.

I have a mac and will try setting it up on MAMP. The MAMP website tries to sell you an upgrade to MAMP Pro which supposedly lets you run multiple servers at the same time. Do you know anything about MAMP, and whether the Pro upgrade is worth it.

Not a clue actually. The only mac I deal with on a regular basis is my iPod Touch.
I'm sure someone else will speak up though.
