Getting this message:
Plugin could not be activated because it triggered a fatal error.
The domain mapping plugin is only compatible with WordPress Multisite.
This is a brand new site install, so it's using the latest versions of everything. Buddypress 1.5.3.1 and Wordpress 3.3.1. Only other plugins installed right now are from WPMU.

I thought I had it set up for multi-sites, but I haven't actually tried to add a second user yet (and that would make sense). This is my second install since Sunday, so it's possible I missed a step the second time around.
Yeah, that's exactly what I did. I set it up right on the test site but didn't turn on MU for the second (real) install. I tested several programs before picking Buddypress so right now they're all starting to blur together.

As it states in your error message:
Its a multisite plugin to map domains to sub sites on a multisite install.

Don't forget to use dedicated ip for domain.
