I've just installed Chat plugin. Then, I've activated it in a test blog (not in the multisite network), in order to make some checks and prepare the spanish translation.
I've got a @Anywhere key from Twitter in order to use Twitter authentication, but when I've tried to put the key in the field, I've got this message:
You do not have sufficient permissions to modify unregistered settings for this site
In fact, I get this message if I do any change in plugin settings.

Hiya,
Not sure why this is happening, but I've asked someone from the dev team to stop by and take a look.
The plugin should be showing the options menu and working correctly for anyone with admin privileges. It also should work fine on WordPress without any sort of network activation.
@blogspnte, finally managed to reproduce the issue. It happens when the plugin is enabled in a Mutisite blog and the user who tries to change the settings doesn't have super admin rights. I'm working on a fix and should have it ready in few hours. I'll upload it here once ready.
Too early to claim victory. Now I can change Chat plugin settings, but I can't get it to work.
I've disabled the bottom corner chat. Then, I've created a page with the chat window, in http://irati.pnte.cfnavarra.es/multiblog/elarequi/chat/. I've set the plugin so that only Twitter authenticated users can chat. Apparently, Twitter authentication works, but sent messages dont' appear in the chat window.
I've tested the chat with four different browsers: IE, Google Chrome, Safari and Firefox.

Anyway, @mohanjith, thank you so much for your effort and patience. Now, I'm tired after a very busy day and I'm going to bed. As we say in Spain, "mañana será otro día" (tomorrow is another day).
We can reproduce the issue on your site but cannot reproduce it anywhere else, can you please verify whether the chat tables are there in the database and whether the messages are in the 'wp_chat_message' (assuming your base prefix is 'wp_')?

I have been able to look at the tables indirectly, by means of the plugin WP-DBManager. There was no 'wp_chat_message' and any other 'xxx_chat_message' table at all. It's obvious that plugin can't write in non existing tables. But why these tables weren't created on plugin activation?
I think that this case has something to do with the initial problem: "Not sufficient permissions to modify Chat plugin settings", because after activating the plugin in the main blog of our multisite installation, two new tables have been created, and so it's possible to chat.
But the problem persists with this other blog: http://irati.pnte.cfnavarra.es/multiblog/elarequi/chat/. Although I've deactivated the plugin and activated again selectively (that's to say, only for this blog), I can't chat.
Probably I could make a massive activation of the plugin to check it, but it doesn't make me happy, because I don't want to create many tables before I've tested and translated the plugin thoroughly.
It works!!! After activating the plugin in the main blog, all the other blogs in the multisite installation seem to be able to chat.
Anyway, there are some minor problems with the look of the writing field, the chat box and some spanish characters in author's names.
I'm going to test this plugin during next week. I'll send you a thorough report, and the spanish translation, of course. By the way, I've noticed that not all the text strings are ready for translation.

Which version are you using and is this user is an admin for the site/blog (not for the network)?

I am using wpmu v 3.0.1
and Date Exclusion SEO v 1.5
this user is an admin for his blog on the network, not a super admin

My bad, I thought the issue was with the Chat plugin.
Please contact the Date Exclusion SEO plugin developer to fix the issue with their plugin.
