Spammers keep coming in even when this plugin is installed. It's like the plugin does not exist at all.
I have searched in Bpress forum and they said this is due to WPMU.
Antispam feature is so important in a cms. Why are they not taking this feature seriously?

check this plugin's description: it says compatible with wordpres multisite 3.1.2
nothing about buddypress, that means its not compatible.

Yeah as Ovidiu says, it's not compatible with BuddyPress at the moment. I'll ask what the developers think about adding BuddyPress support to this one.
Have you tried our anti-splog plugin to help:

And I'd recommend typepad's free solution for comment spam:
http://antispam.typepad.com/info/get-started.html
There's just no way to completely get around it on a site with open registrations, but these two can help significantly.

Thanks for the idea, however this will require lot of changes. We will consider this feature for an upcoming release.

I am generally only getting about 2-3 spam registrations a week now (this is down from about 20 a day).

BuddyPress Moderation is another option for use on budypress enable sites and works network wide.
